A class of Arabidopsis mutants exhibit lesions symptomatic of disease or accelerated cell death. The recessive acd11 mutant, caused by deletion of a sphingolipid trafficking protein, exhibits salicylate-dependent programmed cell death. ACD11 is not an essential gene as removal of SA by the SA hydroxylase NahG, or by mutations in the EDS1 or PAD4 genes required for SA signaling, suppress PCD in acd11. A PCD pathway with similar hormonal and genetic requirements is suppressed by mutations in autophagy (ATG) genes, as atg7 mutants are compromised in the SA-dependent hypersensitive response elicited by the avrRps4 effector, delivered by the bacterial pathogen P. syringae. To better understand such a pathway, suppressors of acd11-related death were isolated by survival after treatment with the non-metabolized SA agonist benzothiadiazole which induces PCD in acd11/nahG. The screens yielded suppressors representing genes in signaling for defense and PCD or effectors of PCD. Complementation analyses sorted alleles, and identified multiple alleles of PAD4 and EDS.
