Genetics has been a cornerstone for plant research; and technological advances in the past 25 years have equipped plant biologists with important genetic tools to facilitate their work. Such advances include methods of plant transformation, strategies for gene tagging and mutagenesis using Agrobacterium and mobile genetic elements, and the ability to knockdown gene expression with RNAi. While powerful, all of these tools have limitations for genetic study: transgenes, for example, integrate randomly on chromosomes and their expression is affected by different chromatin environments; many tools fail in the face of genetic redundancy, and knockdowns provided by RNAi are not complete and subject to epigenetic rules just beginning to be understood. Because of these limitations, plant biologists have long sought-after efficient methods for directly modifying target DNA sequence in vivo (namely gene targeting). In the past about several years, very efficient plant genome modification technology using engineered nucleases has emerged. In this talk, the recent progress and the future challenges in this field will be presented.
