报告摘要:During the development of nervous system, individual neuron needs to extend a growth cone to sense various guidance cues present in the surrounding environment, thus to be guided to its target area and finally connects to its target cell. This dynamic path finding process requires accurate temporal and spatial regulatory machinery. However, the detailed molecular mechanism is not well understood. Here we show that ncal-1, a novel calponin-homology (CH) domain containing protein, functions cell-autonomously in proper formation of neuronal connection. Because CH domain has been implicated in actin association, we are testing the relationship between NCAL-1 and other actin regulatory molecules. Meanwhile, we are also taking immunoprecipitation approaches to identify the NCAL-1 interacting proteins.
(2)报告题目:A negative regulator in RAB-3 hydrolysis

报告摘要:Synaptic vesicle (SV) exocytosis occurs in four consecutive steps: docking, priming, fusion, and recycling. RAB-3 is a SV localized small G-protein of the Rab family. By binding to its downstream effecter RIM (Rab3 interacting molecule), it regulates SV exocytosis at docking and priming processes. During SV fusion with the plasma membrane, GTP bound RAB-3 is hydrolyzed and removed from the membrane to the cytoplasm. Previous studies show that RAB-3-GTP hydrolysis depends on Ca2+ induced membrane fusion. However, how these two events are coupled together is still unknown. Here, we present data to show that a key SV related molecule plays a negative role in RAB-3 hydrolysis.
