Engineering the uORF of TaABA2 Enhances Stress Tolerance and Prevents Preharvest Sprouting in Wheat

    Wheat is a critical staple crop worldwide and frequently experiences multiple abiotic stresses under variable field conditions. The phytohormone abscisic acid (ABA) acts as a core regulator of plant development and stress tolerance. Although ABA metabolism and its downstream signaling cascades have been well characterized, the translational regulatory mechanisms controlling these pathways remain poorly understood. Upstream open reading frames (uORFs) are pivotal translational repressors that modulate protein synthesis of downstream functional genes, representing promising targets for crop genetic improvement. In this study, we focused on ABA2 as a case example. We identified three TaABA2 homeologs in wheat, with TaABA2-5A containing 13 uORFs. We then precisely edited the uATG motif within the uORF of TaABA2 to impair uORF-mediated translational repression. The resulting uorf mutants showed enhanced salt and drought tolerance while suppressed pre-harvest sprouting. Above all, this uORF-engineering approach provides a feasible and promising strategy for the molecular breeding of high-performance, stress-resilient wheat and other cereal crops.
